• Effect of GmLEC1-A Expression on ABA Content at Germination Stage in Soybean(Glycine max)

    LEAFY COTYLEDON1(LEC1) is a key regulator of seed maturation, which gives embryos the ability to withstand desiccation. In this study, a novel transcription factor that is homologous to LEC1 in soybean(Glycine max) was isolated from Dongnong 50 by homologous cloning and was named as GmLEC1-A(GenBank accession number: MF681785). Sequence analysis showed that GmLEC1-A contained conserved B regions, which were functional domains of H4 factor. The relative expression level of GmLEC1-A was the highest in seeds of the soybean cultivar Dongnong 50. To verify the function of GmLEC1-A, ectopic expression of Arabidopsis thaliana and over-expression of soybean plants were generated. In Arabidopsis, the expression of GmLEC1-A restored the tolerance of lec1 mutant to seed drying, indicating that GmLEC1-A was a functional homolog of LEC1,and it might regulate the maturation phase of seed development. In soybean, over-expression of GmLEC1-A caused slower growth and lower germination rates as compared to that of wild-type soybeans. Furthermore, over-expression of GmLEC1-A seemed to increase the level of endogenous abscisic acid(ABA) at the germination stage. These results suggested that GmLEC1-A had a function in regulating ABA content at the germination stage.

  • Proteomic Studies of Petal-specific Proteins in Soybean [Glycine Max(L.)Merr.] Florets

    A survey of petal-specific proteomes of soybean(Glycine max(L.) Merr[Non-italic].) was conducted comparing protein expression profiles in different petals. Two-dimensional polyacrylamide gel electrophoresis reference maps of protein extracts from standard petals(SP), lateral wings(LW), keel petals(KP), and reproductive organs(RO)(a mixture of stamen and carpel) were obtained. Protein expression in the three petal types was compared using Image Master TM 2 D platinum 6.0 software. This indicated that the proportion of homologous proteins between SP and LW was 59.27%, between SP and KP was 61.48%, and between LW and KP was 60.05%. Within a mass range of 6.5-200.0 ku and pH 4.0-7.0, approximately 590, 646, 544, and 700 protein spots were detected in SP, LW, KP, and RO, respectively. A total of 82 differentially expressed proteins were detected. Sixty-four of these detected spots were differentially expressed and showed more than 2-fold changes in abundance; of these 64 proteins, 26 showed increased expression and 38 showed decreased expression. Among these spots, single organ-specific proteins were also identified.They were ID 49(60.9 ku), ID 45(50.0 ku), and ID 46(40.5 ku) in RO, ID 98(42.0 ku) in SP, and ID 05(29.0 ku) in KP. A total of 14 protein spots from 82 differentially expressed proteins were identified with LC-MS/MS. Further protein identification was conducted using the SwissProt and NCBInr databases. The identified proteins and their putative functions were discussed further. This was the first study reporting the comparison of petal protein profiles of soybean florets using proteomics tools.

  • Compatibility Screening of Plant Extracts Synergistic with Osthole

    Plant pests have been a major problem in agricultural production. To establish sustainable methods for plant pest control, 20 of plant material extracts were selected which were synergistic with Osthole to improve the performance of the osthole existing in products. The preliminary screening results indicated that Zanthoxylum bungeanum Maxim.(Z. bungeanum Maxim) compounds exerted the highest synergism. A single dose(2 000 mg · L~(-1)) of Z. bungeanum Maxim. crude extracts was used against Plutella xyllostella(P. xyllostella), Lipaphis erysimi(L. erysimi), and Sitophilus zeamais(S. zeamais). For a further examination form Z. bungeanum Maxim., in which crude extracts were mixed with osthole at different ratios. The volume ratio of 7 : 3(Z. bungeanum Maxim. extract: osthole) exhibited a more pronounced synergistic effect, and all the values of the co-toxicity coefficients were above 120. Eight distinct compounds were derived from the ethanol crude extract from Z. bungeanum Maxim.Then, the toxicity of these components to pests, such as P. xyllostella, L. erysimi, S. zeamais, Rhizopertha dominica(R. dominica),and Tribolium castaneum Herbst(T. castaneum Herbst), was determined. The results indicated that Compound 4 and Compound 7 exerted lethal effects on pest investigation. Compound 4 had the most substantial insecticidal action, at a concentration of64 ug · mL~(-1), and a death ratio of 78.3% was achieved for P. xyllostella.

  • Bioinformatics and Expression Pattern Analysis of Tomato ns LTP 2-like cDNA full-length Gene Clone

    TDF1(transcription-drived fragment) was homologous to the predicted S. lycopersicum nonspecific lipid-transfer protein,nsLTP 2-like(91%), and it was significantly upregulated in response to C. fulvum(cladosporium fulvum) infection in tomato plants.In this experiment, the full-length cDNA of nsLTP 2-like was cloned using RACE technology based on the sequence of TDF1(GenBank: JZ717725). A full-length, 625 bp(GenBank: KU366289), cDNA sequence, which with 98% similarity to nsLTP 2-like gene(GenBank: XM015233692) was obtained. This cDNA contains an ORF(open reading frame) with full-length of 345 bp, coding of 114 amino acids, including 12.3% Ala and Gly. Protein molecular weight was 11.51 ku, the isoelectric point(pI) was 8.99, and average overall hydrophilicity was 0.412, with one phosphorylation sites, belonging to volatile acidic nuclear protein. Secondary structure prediction showed that α-Helix accounts for 30.7%, extension chain for 12.28%, β-corner for 9.65%, and random coil for 47.37%. Through comparative analysis of the homology among species, it was found that the amino acid sequence of tomato nsLTP 2-like protein had a high similarity with other plants, and with a specific conserved sequence which might related features in nsLTP 2-like protein. It also be analyzed the gene expression pattern of tomato in different parts and under different stress conditions.The results showed that nsLTP 2-like gene was up-regulated in varying degrees, under the condition of cold stress, exogenous hormone spraying and cladosporium fulvum infection. Therefore, it was speculated that the gene played a role in response to abiotic and biotic stress in tomato.

  • Molecular Differentiation of Different Pathogenic Phenotypes of Infectious Bursal Disease Viruses by RT-PCR Combined with Restriction Fragment Length Polymorphism(RFLP) Assay

    Accurate differentiation of the pathogenic phenotypes of infectious bursal disease viruses(IBDVs) will instruct effective vaccination programs and improve the study of the molecular epidemiology of IBDVs. In this study, an 833 bp hypervariable nucleotide region was identified in VP2 genes of known IBDVs with different virulences through multiple sequence alignment.Moreover, using NEBcutter software analysis, two restriction enzyme sites, SpeⅠ(generating 531 and 302 bp fragments) and StuⅠ(generating 242 and 591 bp fragments) were found presented in very virulent but not attenuated IBDVs. Moreover, the restriction enzyme site SacⅠ(generating 218 and 615 bp fragments) presented in attenuated IBDVs but not very virulent IBDVs. Therefore,a reverse-transcription(RT)-PCR combined with a restriction fragment length polymorphism(RFLP) assay was developed to differentiate attenuated and very virulent IBDVs. The RT-PCR assay was used to confirm 282 IBDV positive samples from 310 suspicious dead chicken samples. The 60 IBDV positive samples were used to evaluate the assay, followed by confirmation via gene sequencing and histopathological examinations of the bursas of Fabricius from chickens infected by these IBDVs. The results showed that 24 viral strains with SpeⅠand StuⅠsites were very virulent, causing severe pathological damage in the bursas of Fabricius, while36 viral strains with the SacⅠsite were attenuated IBDVs, exhibiting only slight pathological damage. The combined RT-PCR and RFLP assay provided a useful approach for differentiating the pathogenic phenotypes of IBDVs.

  • Effect of Xylazine Anesthesia on Glu and GABA Amino Acid Neurotransmitters in Rat

    Neurotransmitters of the central nervous system were the important way to study the mechanism of anesthesia. The effect of different doses of xylazine anesthetic on the glutamate(Glu) and γ-aminobutyric-acid(GABA) were investigated and the mechanism of xylazine anesthetic on the central nervous system were explored in this study. A total of 88 rats were randomly divided into three groups, including normal saline control group, group with low dose of xylazine and group with high dose of xylazine.Cerebrum, cerebellum, hippocampus, thalamus and brainstem were collected. The results showed that the concentration of Glu in the hippocampus, thalamus and brainstem decreased first and then increased, but it increased first and then decreased in the cerebrum and cerebellum during the period of anesthesia. The concentration of GABA in the cerebrum, thalamus, brainstem and hippocampus increased first and then decreased. The results showed that xylazine inhibited Glu and promoted GABA with different dose dependence. The results and methods could provide guides for the clinical use of xylazine.

  • Chlorophyll Content Retrieval of Rice Canopy with Multi-spectral Inversion Based on LS-SVR Algorithm

    To monitor growth and predict the yield of rice over a large area, the chlorophyll contents in the rice canopy were estimated using the unmanned aerial vehicle(UAV) remote sensing technology. In this work, multi-spectral image information of the rice crop was obtained using a 6-channel multi-spectral camera mounted on a fixed wing UAV, which was flown 600 m above the ground, between 11: 00-14: 00 on a sunny day in summer. The measured chlorophyll values were collected as sample sets. The s-REP index was screened out to estimate chlorophyll contents through the analysis of six kinds of spectral indexes of chlorophyll estimated capacity. An inversion model of the chlorophyll contents was then built using the least square support vector regression(LS-SVR)algorithm, with calibration and prediction R-square values of 0.89 and 0.83, respectively. Finally, remote sensing mapping for a UAV image of the Fangzheng County Dexter Rice Planting Park was accomplished using the inversion model. The inversion and measured values were then compared using regression fitting. R-square and root-mean-square error of the fitting model were 0.79 and 2.39,respectively. The results demonstrated that accurate estimation of rice-canopy chlorophyll contents was feasible using the LS-SVR inversion model developed using the s-REP vegetation index.

  • Using Deep Learning for Soybean Pest and Disease Classification in Farmland

    To accurately identify soybean pests and diseases, in this paper, a kind of deep convolution network model was used to determine whether or not a soybean crop possessed pests and diseases. The proposed deep convolution network could learn the highdimensional feature representation of images by using their depth. An inception module was used to construct a neural network. In the inception module, multiscale convolution kernels were used to extract the distributed characteristics of soybean pests and diseases at different scales and to perform cascade fusion. The model then trained the SoftMax classifier in a uniformed framework. This realized the model of soybean pests and diseases so as to verify the effectiveness of this method. In this study, 800 images of soybean leaf images were taken as the experimental objects. Of these 800 images, 400 were selected for network training, and the remaining 400 images were used for the network test. Furthermore, the classical convolutional neural network was optimized. The accuracies before and after optimization were 96.25% and 95.81%, respectively, in terms of extracting image features. This type of research might be applied to achieve a degree of automation in agricultural field management.

  • Study on Multi-variables Decoupled Fuzzy Controller for Confined Pig House in Northern China

    In winter, the confined pig house of northern China is severe. The environment variables are nonlinear, time-varying and coupled, which seriously affect the health of pigs and the qualities of the meat. In order to solve the problem multi-variables coupling, a multi-variables decoupled fuzzy logic control method was proposed. Two fuzzy logic controllers were designed based on fuzzy logic theory. The fans, heaters and humidifiers were used to control temperature, humidity and ammonia. The reductions of temperature and humidity caused by ventilating were compensated by heaters and humidifiers respectively which realized the multivariables decoupling. The proposed methods were validated through theoretical, experimental and simulation analysis. The results suggested that the methods were able to regulate the confined pig house environment effectively. In addition, comparing to the manual regulation, the proposed methods could reduce 19% power consumption as well.

  • Analyzing Factors Contributing to Farmers' Willingness to Participate in Rural Cooperative Finance Organizations

    It has been commonly recognized that the cooperative finance organizations in China's rural areas have been playing an important role in spreading farm production risks, increasing farmers' income, as well as stimulating the overall rural economic growth. This paper attempted to identify the key factors which potentially contributed to the farmers' willingness to participate in the new-form of the rural cooperative finance organizations sponsored by the Chinese governments. In the process, a total of 442 farmer households as well as small-micro farming enterprises were sampled from the villages in Panjin municipality of Liaoning Province in China, where the new form of the rural cooperative finance organization(NFRCFO) experiment was under way. Those potential factors were classified into the four categories, including the characteristics of the famer households, the reputation of the rural finance cooperatives, transaction costs paid, as well as service quality. A discrete Logit model was used for parameter estimations. The results showed that most of the factors being considered exhibited statistically significant effects on the farmers' willingness to take part in NFRCFO, but with each presenting different levels of impacts. The causes and effects for those uncovered empirical results were fully discussed following by policy implication discussion.
