• Strain Cultivation Stage and Infectious Concentration on Soybean Cotyledonary Node via Agrobacterium–mediated Transformation System

    In soybean cotyledonary transformation,Agrobacterium infection is influenced by many factors.The cultivation stage of Agrobacterium strain and infectious concentration were studied in this research.Results showed that the highest rate of resistant shoots was obtained when the strain cultivation concentration was OD 600 =0.6,and the infectious concentration was OD 600 =0.5.Seven soybean cultivars in North China were also screened for high susceptibility to Agrobacterium and three genotypes were found to be suitable for transformation.This research has built a good foundation for soybean transformation improvement.

  • Analysis on Genotypic Differentiation of Phytophthora infestans by Using Random Amplified Polymorphic DNA (RAPD)

    A total of 104 isolates including two Korean isolates and three Japanese isolates of Phytophthora infestans collected from Heilongjiang and Jilin Provinces from 2006 to 2008 were used to determine their mating types,metalaxyl resistance,and RAPD genotypes.All the isolates of P.infestans collected from Heilongjiang and Jilin Provinces belonged to A 1 mating type,and no A 2 mating type was detected.Frequencies of metalaxyl resistant isolates were 94.4%,47.8% and 75.0% in 2006,2007 and 2008,respectively.According to RAPD analysis by using OPD-20 primer,104 isolates including two Korean isolates and three Japanese isolates were separated into 10 RAPD groups at 90.8% similarity level.A total of 10 RAPD groups were not correlated to groups defined by mating type,locations,and metalaxyl resistance.

  • Chlorpyrifos Detection by Piezoelectric Biosensor Based on Acetylcholinesterase Immobilization

    Acetylcholinesterase (AChE) was immobilized on multilayer films assembled by poly diallyldimethylammonium chloride (PDDA) and ι-carrageenan (IC) on silver-coated crystal electrode surfaces to detect the chlorpyrifos belonging to the organophosphates pesticide.Mass sensitive quartz crystal microbalance (QCM) was used to study the effect of AChE concentration and pH of phosphate buffer solution on immobilized acetylcholinesterase.The optimized conditions were as follows: pH was 6.0 which was near isoelectric point (pI);enzyme concentration was 0.1 mg·mL -1;and the lowest concentration of chlorpyrifos that could be detected was 0.1 μg·L -1 .

  • ISSR Analysis of M_1 Generation of Gladiolus hybridus Hort Treated by EMS

    Gladiolus hybridus Hort is one of famous cutting flowers,being famous of big size,bright color,various shapes and long bloom period.New species should be cultivated in order to meet consumers' needs.Mutagenic breeding is a shortcut to cultivate new species of flowers.In this study,corm bud of G.hybridus Hort was treated with different concentrations of EMS.Then M 1 generation was analyzed by ISSR.Results showed that EMS was a very effective mutagenic agent for the corm bud of G.hybridus Hort.With the increase of EMS concentration,the mutagenic rate was increased,among which 1.0% EMS treatment had the biggest influence.However,with the same EMS concentration,there is no close relationship between the amount of mutagenic agent and the divergence of plants genomes,which offered a molecular basis for selecting plants with good mutation.

  • Modification of Cre Gene by PCR Method

    Cre/LoxP site-specified recombination system is mainly used for excision,inversion and integration of target gene.Therefore,this system can be used for plant marker free genetic transformation,site-specific transgene expression and so on.However,the application of this system was limited due to its low expression and excision efficiency.In this study,an intron,which can enhance gene expression in plants,was inserted into Cre by using PCR method.And a modified Cre gene,named Crein,was obtained.This gene was composed of three parts: ATG,intron,Cre gene without ATG.This gene can be used for further research to provide the good system for plant marker free transformation.

  • Isolation of a Bacterium Strain Degraded Agar

    One in 58 strains of bacteria isolated from the compost showed clear colonies after a few days of growth on the plates containing medium made of only agar and water.Water suspension contained only agar (2 and 8g·L -1 ) with two controls (normal saline,LB medium) was inoculated with the bacterium BR5-1 to see whether there was an increasement of the alive bacteria concentration after 48 h of the growth.The results showed that there was a significant rising of the alive bacteria concentration in the agar suspension (6.5×10 3 and 6.0×10 3 cfu·mL -1 ) compared to that in the normal saline control (6.1×10 2 cfu·mL -1 ),and it was far lower than that in LB (Luria-Bertani) control (1.1×10 8 cfu·mL -1 ).In conclusion,the bacterium strain BR5-1 could degrade agar.

  • Water Consumption and Use Efficiency of Main Crops on Semi-drought Area of Western Liaoning Province

    The laws of water consumption in corn,peanuts and millet on the semi-drought area of western Liaoning Province were studied through the FAO-Penman Monteith method and the water balance method.Among three corps,the amount of the day water demand,the whole growth period water demand and the soil water deficit of corn were all the largest.At the same time,its degree of agreement between the water demand and the level of precipitation was the worst,and its average in crop coefficient was larger.The amount of the day water demand,the soil water deficit and its average in crop coefficient of millet were larger.Because of its short growth period,the whole millet growth period water demand was the least,resulting in its degree of agreement between the water demand and the level of precipitation was better.For peanuts,the amount of the day water demand,the soil water deficit and its average in crop coefficient were both the least,the whole growth period water demand was less,degree of agreement between the water demand and the level of precipitation was better,the average in crop coefficient was less,the change of crop coefficient was relatively steady,and water demand in the early and middle period of crop growth was less.From water use efficiency,crop plant order of regional priorities should be corn,millet,and peanuts.From economic benefits of water,crop plant order of regional priorities should be peanuts,corn,and millet;and crop plant order of priority to irrigation should be peanuts,millet,and corn.

  • Evaluation on Heavy Metals in Black Soils of Hailun and Bayan Counties in Northeast China

    The concentration of heavy metals was investigated in black soil samples collected from the fields treated with chemical in Bayan County which were from the vegetation covers,bare soil and grain fields without any chemical treatments in Hailun National Field Research Station of Agroecosystem,Chinese Academy of Sciences (CAS) in July,2009.Results showed that the presence of significantly high concentration of zinc (Zn) and lead (Pb) in soils covered with grass vegetation.However,mercury (Hg),copper (Cu),nickel (Ni),cadmium (Cd),and arsenic (As) were found within the permissible limit set by international environmental agencies.

  • Physiology and Biochemistry Changes of Euphorbia pulcherrima During Leaf Color Transformation

    Reasons for the color change of E.pulcherrima were studied with physiological and biochemical paraments such as: chlorophyll,carotenoids,anthocyanin,Phenylalanine ammonia-lyase (PAL),pH of cell sap in leaves and soluble sugar content etc.The results showed that during the period of the leaf color transmittion of E.pulcherrima,the contents of plastid pigment,soluble sugar and pH of cells sap in leaves showed a high-low-high dynamic change,while the contents of anthocyanin and PAL activity showed the low-high-low dynamic change.Anthocyanin stability had the close relationship with pH of cell sap in leaves,temperature and other conditions.

  • Genetic Diversity Analysis of Aneurolepidium chinense (Trin) Kitag by ISSR Marker

    In the study,the reasonable sampling of the grey Aneurolepidium chinense of green grassland,the grey-green A.chinense of green grassland,the grey A.chinense of Wulimu and the grey-green A.chinense of Wulimu were analyzed by ISSR.Eight primers with clear and diverse products were screened out from 20 primers and 47 DNA fragments were amplified from 39 individuals.The average number of DNA fragments produced by each primer was 5.9,and polymorphic bands were 41 and the polymorphic rate was 87.23%,which could reflect the genetic variation that was with four plots of each kind and to the individuals.Cluster results showed that at first,the grey A.chinense of Wulimu and the grey-green A.chinense of Wulimu were grouped together,and then they grouped with the grey A.chinense of green grassland into one cluster,at last with the grey-green A.chinense of green grassland.A.chinense of identical synusiologic type was grouped together.According to Shannon index,the green grassland of the grey A.chinense had the largest variation degree,which indicated that A.chinense had variation among species and interspecies by ISSR analysis,but A.chinense didn't differentiate different species.

  • Effect of Functional Compound Recipe Liquid of Eliminating Lead on Eliminating Lead and Calcium in Rats

    The experiment was conducted to approve the efficacy of functional compound recipe liquid of eliminating lead and calcium to eliminate lead in rats.Total 120 rats were divided into six groups according to weight: blank group (CK),negative group (ND),positive group (PD) and high group (HD,6.67 Mg·kg -1·BW·d -1 ),middle group (MD,3.33 Mg·kg -1·BW·d -1 ) and low group (LD,1.67 Mg·kg -1·BW·d -1 three groups).Except CK,other groups were drunk 0.24% Pb(AC) 2 ,in which rats were filled with functional compound recipe liquid to eliminate lead and DSMA was added to positive group (PD).On the 20th day,the blood was collected from the tail.on the 40th day,the whole blood was collected from eyeball.After executed,liver and blood were collected to determine the content of lead and calcium.Results showed that lead contents in blood and liver of three dosage groups obviously decreased compared with ND and there was significant difference between any two groups of three dosage groups (P<0.01).Meanwhile,the calcium concentration increased in three dosage groups.It is concluded that the functional compound recipe liquid of eliminating lead has the evident function of eliminating lead and can promote calcium absorption.

  • Evaluation on Environment State of Breeding Area in Shuidong Bay by Using Fuzzy Comprehensive Evaluation Method

    Fuzzy comprehensive evaluation method was used to develop a standard model to analyze and evaluate nearness degree of water environment quality at breeding area of Shuidong Bay in China.Results showed that certain environment contamination factors in some areas seriously exceeded the standard value and led to the whole water quality at the third class level.The measurements should be taken to promote the sustainable development of breeding area in Shuidong Bay.

  • Promotion of Agricultural Products Competitive Power in Heilongjiang Province with Modern Logistics

    The paper studied the competitive power of agricultural products in Heilongjiang Province by analyzing the revealed comparative advantage,and drew the conclution that agricultural products' competitive power was decreasing in Heilongjiang Province.There were three main reasons for this status: firstly,the cost of circulation of agricultural products was increasing;secondly,the commodity rate of agricultural products was low;thirdly,the marketing power of agricultural product was weak because of lacking the macro-management mechanism.To solve this problem,modern logistics must be developed to promote the competitive power of agricultural products in Heilongjiang Province.
